| Resumen | Cookeina speciosa (Fr.) Dennis is a fungus belonging to the phylum Ascomycota, subphylum Pezizomycotina, class Pezizomycetes, order Pezizales, family Sarcoscyphaceae, genus Cookeina. This genus is characterized by its cup-shaped stipitate apothecia, with bright colors ranging from red to orange, due to their high carotenoid pigment content; with fasciculated excipular hairs or [1, 2]. They are saprobic, meaning they degrade organic matter, mainly woody substrates. They are distributed in temperate regions of the northern hemisphere and tropical regions of the southern hemisphere, where they are most abundant. Globally, they have been reported in Africa, Asia, and Australia, while in Mexico they have been recorded in the states of Tamaulipas, Quintana Roo, Chiapas, Nayarit, and Oaxaca, where they grow in groups during the rainy season [1]. However, there are still no detailed taxonomic studies on this genus for Mexico, and C. speciosa in particular has antimicrobial properties due to its secondary metabolites [3], making it a fungus with great potential. Morphological characteristics such as color, size, shape, and surface of the ascoma wall were observed using a stereoscopic microscope (Leica model: S9-E). In addition, a scanning electron microscope was used to observe the surface details of the peridium with better resolution. This process was carried out by coating a thin layer of gold on a sample of the dry biological material in order to avoid charging under the electron beam. Longitudinal sections of the ascoma were made to observe the internal structures under a compound microscope (Olympus CX3). The sample was prepared beforehand by rehydrating it with 70% alcohol and adding 10% potassium hydroxide (KOH) to clarify the structures and allow form better visualization. Microscopic description: Apothecium 100-500 x 100-800 μm, growing solitary or gregarious, centrally stipitate, globose or deeply or shallowly cupulate, orange to yellow when fresh; stipe 30-750 x 10-60 μm, thin, hollow, slightly attenuated below and often forming a disc-shaped attachment at the bottom, concolorous or even paler than the receptacle, margin with three rows of white hairs; hairs up to 200-250 × 150-190 µm, conical in shape, grouped in bundles of parallel hyphae arising from the periphery of the ectal excipulum; Two-layered ectal excipulum, outer ectal excipulum 28-80 μm, composed of thick-walled, hyaline to light brown cells, globose to angularis or prismatic in texture, arranged perpendicular to the surface of the receptacle; internal ectal excipulum 30-300 μm, composed of cells with an intricate texture, the layer is gelatinized only in young fertile specimens, disappearing at maturity. The medullary excipulum is composed of cells with an intricate to porrect texture; subhymenium 15-30 μm, composed of cells with an intricate to porrect texture, gelatinized in immature specimens. Hymenium surface smooth, orange-yellow, grayish brown to pale chocolate brown, very hygrophane, becoming uniformly concolored when dry, pastel yellow to brownish yellow; Paraphyses 2-4 µm wide, numerous, filiform, highly branched at the apex, slightly protruding from the asci. Asci 300-370 × 17-19 μm, unitunicate, operculate, non-amyloid, octosporous. Ascospores 22-33 × 10-16 μm, ellipsoid, thick-walled, bigutulate, in SEM, ornamentation with longitudinally undulate striations, some anastomosed, not parallel [4]. Cookeina speciosa is a species with distinctive and well-defined morphology that allows it to be identified and distinguished from other genera within the family Sarcoscyphaceae. Analysis using optical microscopy allowed for detailed documentation of the internal organization of the apothecium, while scanning electron microscopy provided high-resolution ultrastructural information, allowing for detailed observation of the ornamentation of the ascospores, a diagnostic character relevant for the taxonomic identification of the species. The record of C. speciosa in Mexico expands knowledge about the distribution and diversity of the genus Cookeina in the country, where detailed taxonomic studies on this family remain scarce; therefore, it is necessary to implement SEM inm the observation of taxonomic characters that facilitate its diagnosis.
